The Effect of Insulin and Deprivation of Food on Metabolism of Acetoacetic Acid by Muscle.

چکیده
Previously our laboratory demonstrated that voluntary muscle fiber groups and diaphragms from depanereatized and alloxan-diabetic rats take up leas acetoacetic acid than fiber groups and diaphragms from control animals (1, 2). The addition of glucose and insulin causes an increase in acetoacetic acid uptake by muscle fiber groups from both control and depancreatized rats. These experiments were performed on muscle from both fed rats and rats that had been deprived of food for 24 hours. It was concluded that the decrease in ketone body utilization by muscle from insulin-deficient animals is a contributing factor in diabetic ketosis. These conclusions have been further substantiated by Scow and Chernick (3), who reported that the utilization of both acetoacetic and /3-hydroxybutyric acid is reduced in insulin-deficient rats. &reeve (4) followed the conversion of acetate-l-% to ketone bodies in diabetic patients and also presented evidence in support of impaired utilization of P-hydroxybutyric acid. It has been demonstrated, by measuring patterns of enzyme activity, that the pathways by which the major portion of the ketone bodies are produced in liver differ in the ketosis associated with deprivation of food and that found in diabetic rats (5). The present paper reports an investigation of the effect of prolonged deprivation of food on the uptake of acetoacetic acid by diaphragms and skeletal muscle fiber groups of rats and the effect of insulin’ on the uptake of acetoacetate-3-14C2 and its conversion to l-4C02 by these muscles.

Acetoacetic acid metabolism by skeletal muscle fibers from control and diabetic rats.
Previously we demonstrated that both diaphragm and adductor fibers from alloxan diabetic rats take up less acetoacetic acid than diaphragm and fibers from control rats (1). To date it has not been possible to show an influence of insulin on ketone utilization by muscle (2). The present paper extends the previous Fork on acetoacetic acid uptake to pancreatectomized rats, which show a decrease in...
متن کاملEffect of insulin on carbohydrate metabolism of skeletal muscle fibers and diaphragm from control and pancreatectomized rats.
Rat diaphragm muscle has been used almost exclusively for the investigation in vitro of skeletal muscle metabolism. However, the diaphragm is a specialized muscle adapted to constant activity and, with respect to oxygen consumption (1) and oxidative metabolism, seems to occupy an intermediate position between constantly active heart muscle and intermittently active skeletal muscle (2, 3). Histo...

متن کاملEffect of Eight Weeks of High Intensity Interval Training on Insulin Resistance and IRS1 Gene Expression in Gastrocnemius Muscle of Obese Wistar Rats
Background and objectives: The role of genetic components in expression of proteins involved in signaling pathways of fat and carbohydrate metabolism has been well-demonstrated. The aim of this study was to determine effects of high intensity interval training (HIIT) on glucose, insulin, and insulin resistance levels as well as IRS1 expression in gastrocnemius muscle of obese Wistar rats. Meth...
متن کاملEffect of stress due to food deprivation, social inequality and instability on brain
Introduction: Several reports show a relationship between health equity, and health status and longevity of individuals. In this study, effects of food deprivation, inequality and social instability were investigated on the formation of lipofuscin pigment in Balb/C mice by histopathological and biochemical methods. Methods: Thirty-six male mice were divided into 6 groups, then different kind...
